BHB-Phe, a naturally occurring body compound, influences appetite and body weight by activating specific brain cells.

Researchers from Baylor College of Medicine, Stanford University School of Medicine, and other institutions have identified a newly discovered metabolite: BHB-Phe. This naturally occurring substance has a direct impact on the brain. According to a recent publication in the leading journal Cell, BHB-Phe regulates both appetite and body weight through interaction with neurons.

BHB, short for beta-hydroxybutyrate, was already known as an alternative fuel source produced by the liver during fasting or intense physical activity. But this discovery sheds new light on its role in the brain — and opens possibilities for treating obesity.

An Unknown Link in Metabolism

The team led by Dr. Jonathan Z. Long (Stanford) discovered that the enzyme CNDP2 plays a key role: it couples BHB with amino acids, with BHB-Phe being the most common. Animal studies show that BHB-Phe affects body weight and metabolism.

Dr. Yong Xu (Baylor) investigated the effect of BHB-Phe on mice. The results showed that the compound activates specific neurons in the hypothalamus and brainstem — brain regions involved in appetite regulation. In mice unable to produce BHB-Phe, food intake increased and weight gain occurred.

Think of amino acids as letters in the alphabet of life. Just as letters form words, amino acids form chains that create proteins. These proteins are the ‘words’ that determine how your body functions and grows.

BHB-Phe versus Lac-Phe: Similarities with Nuance

The same enzyme CNDP2 also produces Lac-Phe — a related compound released during exercise. Earlier studies, published in Nature, showed that Lac-Phe also suppresses food intake in mice. Yet both compounds appear to achieve their effects through different pathways.

Analysis of brain activity revealed that the majority of activated neurons are unique to each compound. An unexpected finding — suggesting that the brain uses multiple, partially separate systems to regulate appetite. The fact that BHB-Phe also occurs in humans and may be disrupted in obesity makes the subject even more urgent.

“This research opens many new possibilities,” says Long. “Perhaps we can regulate appetite with BHB-Phe without the need for strict diets or low-carbohydrate eating patterns.”
